Researchers have identified the distinctive compound behind olive oil's broad spectrum of wellness and longevity-promoting effects. Designated oleuropein, this polyphenol can contribute to reduced LDL cholesterol and hypertension, cancer prevention, defense against oxidative harm, and protection from cognitive deterioration.1,2 Oleuropein delivers the characteristic sharp, peppery, somewhat acrid taste found in premium extra virgin olive oils.2 It also accounts for the majority of olive oil's antioxidant, anti-inflammatory, and disease-combating properties.2-4 Notably, when oleuropein was administered to tumor-bearing animals, the masses entirely receded and vanished within 9 to 12 days!5

Hypertension

Experiments in animals indicate that olive leaf extracts produce considerable reductions in elevated blood pressure readings.6,7 Strikingly, these outcomes appear whether the supplementation begins prior to or following the onset of hypertension. This demonstrates that the extracts possess both preventive and therapeutic value for high blood pressure.8

The pressure reduction coincides with diminished strain in the heart's left ventricular chamber. This enhances circulation through the coronary arteries supplying the heart muscle. Further human trials confirm the capacity of olive leaf extracts to meaningfully lower blood pressure values.9

A clinical trial compared olive leaf extract against captopril, a standard pharmaceutical for managing hypertension.11 In this investigation, individuals with stage-1 hypertension (140-159 mmHg over 90-99 mmHg) received either 500 mg of olive leaf extract twice daily, or 12.5 mg of captopril twice daily, with the latter titrated up to 25 mg twice daily when warranted. After 8 weeks of therapy, both cohorts showed declines from their initial mean blood pressure (11.5 and 13.7 mmHg systolic; 4.8 and 6.4 mmHg diastolic, respectively), with no statistically meaningful difference between groups. They elevate nitric oxide synthesis, a signaling compound that facilitates vessel relaxation.16,17 They curtail the generation and function of a group of compounds called matrix metalloproteinases, or MMPs.18–20 Excessive MMP activity essentially degrades the gel-like scaffolding binding cells together, rendering vessel linings more susceptible to plaque injury. They also shield LDL-cholesterol from oxidation, one of the earliest events in atherosclerotic development.21–23 Oxidized LDL provokes inflammation, further harming arteries, and olive leaf extract provides multi-targeted anti-inflammatory action.20,24,25

Polyphenolic substances present in olive foliage have demonstrated direct ability to prevent arterial plaque formation (and consequently diminish the likelihood of cardiac and cerebrovascular events) via two pathways. Initially, they decrease the generation and function of certain "adhesion molecules."26–28 These compounds prompt white blood cells and platelets to adhere to arterial walls, initiating early plaque development. Additionally, they diminish platelet aggregation (clumping) through various mechanisms, lowering the probability that small clots will develop at plaque sites to trigger a stroke or cardiac event.29,30

Blood Sugar Disorders

The diabetic (and pre-diabetic) condition of persistent blood sugar elevation generates considerable oxidative stress throughout the organism, sparking inflammation and tissue injury that dramatically hastens aging. Approaches to diabetes management pursue two primary objectives:

  1. restoring blood glucose to normal ranges, and
  2. minimizing the harm inflicted by unavoidable blood sugar fluctuations that still arise.

Olive leaf extracts are demonstrating genuine potential in both domains. In animal and laboratory investigations, olive leaf extracts and oleuropein have been observed to reduce blood sugar through multiple pathways.31–34 They retard the breakdown of starches into simple sugars, slow intestinal absorption of those sugars, and enhance glucose uptake into tissues from circulation.31,32 They shield tissues from oxidative injury caused when glucose attaches to proteins through a process termed glycation.33,34 They additionally elevate levels of other intrinsic antioxidant defenses within the organism, extending the protective scope.33

These pathways yield directly measurable advantages. Studies reveal that diabetic animals given olive leaf extracts experience significant decreases in blood sugar and cholesterol values.33,35 In a striking head-to-head comparison, diabetic rats received either olive leaf extract or glyburide (Diabeta®), a widely prescribed glucose-lowering pharmaceutical.35 By the study's conclusion, the antidiabetic properties of the extract exceeded those of the medication.35

One compelling study demonstrated that when laboratory rats consumed a high-fat, high-carbohydrate diet, they developed every hallmark of metabolic syndrome (excess abdominal adiposity, hypertension, unfavorable lipid patterns, and compromised glucose tolerance).36 However, when animals received that detrimental diet alongside olive leaf extracts, nearly all metabolic disturbances improved or, in certain instances, normalized entirely.36

Clinical investigations reveal that supplementing with 500 mg of olive leaf extract once daily produced meaningful decreases in hemoglobin A1c values, the standard indicator of chronic exposure to elevated blood sugar among diabetic individuals.31 Supplementation additionally reduced fasting plasma insulin levels, a significant finding given that chronic insulin elevation may contribute to the elevated cancer risk observed in diabetics.31,37

Malignancy

The Mediterranean dietary pattern is celebrated for its capacity to lower cancer risk.38,39 While multiple components of this diet contribute to risk reduction, accumulating evidence indicates that olive oil—and specifically its oleuropein content—represents crucial elements of the diet's cancer-protective properties.

Oleuropein and olive leaf extracts possess numerous additional anticancer mechanisms:

  • They help suppress inflammation, another significant promoter of tumor proliferation.41
  • In breast cancer cells specifically, oleuropein diminishes malignant cells' capacity to respond to estrogen, the female hormone upon which many breast cancer cells depend for survival.43
  • Oleuropein blocks the generation of the "protein-dissolving" enzymes that cancer cells require to penetrate healthy tissues and spread to distant sites throughout the body.19

These mechanisms have now been demonstrated in laboratory and animal studies to reduce both occurrence rates and subsequent progression of a broad spectrum of cancers, including those affecting the brain, head and neck, breast, liver, bladder, prostate, and skin, along with leukemia.42,43,45–49

In one particularly striking investigation, mice with elevated spontaneous cancer rates received oral oleuropein supplementation.5

The tumors entirely receded and vanished within 9 to 12 days! When examined before they disappeared, the tumors displayed a disordered, crumbly texture, and no viable cancer cells remained within.

Neurological Defense

Olive extracts assist in safeguarding the brain and central nervous system from the devastation caused by strokes and age-related degenerative conditions such as Alzheimer's and Parkinson's diseases.50 They achieve this through inflammation suppression and reduction of oxidative damage.

In acute brain injuries such as those triggered by stroke or trauma, destructive processes including oxidative stress manifest within minutes of the initial event—and, paradoxically, are exacerbated by the restoration of normal circulation to the affected area.50,51

Olive leaf extracts provide analogous protection against neurodegenerative disorders. Oxidative stress develops more gradually in neurodegenerative conditions. Nevertheless, the effects accumulate over time, generating inflammation and other alterations that lead to buildup of abnormal proteins that disrupt brain function and destroy neurons. Through a mechanism identical to allopurinol (the premier pharmaceutical therapy for gout), oleuropein prevents uric acid buildup by blocking xanthine oxidase, the enzyme that transforms DNA and RNA into uric acid.57

Oleuropein has additionally been demonstrated to assist in preventing and addressing symptoms of rheumatoid arthritis. When administered at the earliest indication of arthritis in animal models, oleuropein prevented symptom development and also produced notable improvement in the microscopic appearance of joint tissue from affected animals. When administered after arthritis had fully developed, significant improvement in inflammatory changes to joints was observed, compared to untreated animals.58

Oleuropein produced comparable benefits for osteoarthritis. In animal models of this degenerative joint condition, olive leaf extract reduced joint swelling, improved the microscopic appearance of joint tissue, and prevented the production of inflammatory cytokines.59
